spamassassin, clamav etc

Hi, Its been years since I have had any real interaction with these programs but I have a quick question
I have installed the whole gammit of mail service on my vestacp server and I have a few email accounts set up as well. I find I am getting spam lol and have no idea why I should be, but any whats the internet without spam. I utilise roundcube to access my emails (they are forwarded to centralise email account). My quiestion I guess is if I consign spam the roundcube junk mail folder does that information end up being known to and acted apon by spamassassin so therefore endup being blocked in the future.
